工作原理
扫描近场光学显微镜是一种基于扫描探针技术的具有高分辨率和荧光标记特性的,可直接观察微观领域的显微成像设备。当光波照射物体之后,光波可以分为近场波及远场波,远场波由于受传播的衍射极限限制,不能实现高分辨率的成像,这也是普通光学显微镜成像分辨率受限的原因,近场波不受衍射极限限制,但随距离逐渐衰减,所以需要在物体的表面近距离收集近场波信息。为实现这一目的,扫描探针技术的基本原理是:一个亚波长孔径的微小光源,在一物体的近场范围内照射物体时,照射光斑的面积只和孔径大小有关,而与波长无关。这样,在反射光或透射光中,将携带物体亚波长尺寸结构的信息,通过扫描采集样品各“点”的信号光,即可得到分辨率小于半波长的样品的近场图象。基于这一原理,扫描近场光学显微镜成像过程是:主机通过样品台对样品进行固定并进行光路对准,然后通过外置激光光源(另行购买,可以根据测量需要,自己选择多种波长匹配)将激光通过光纤耦合器提供给光纤探针;光纤探针固定在控制箱的压电陶瓷音叉上,平行于样品表面振动,音叉通过电压的变化监控样品与探针的距离,使其保持不变,回馈电信号的大小变化就反映了样品表面的高度变化,以此得到样品表面的形貌图,实现高精度三维扫描。校准光栅又称为光栅尺位移传感器,是利用光栅的光学原理工作的测量反馈装置,可用作直线位移或者角位移的检测,其测量输出的信号为数字脉冲,具有检测范围大,检测精度高,响应速度快的特点,在本仪器中实现光纤探针在物体表面(纳米尺度)的高速精确定位。

2、学科专业现状
生物系现有副教授1名,博士5人,博士后2名,硕士4名,兼职教授3名,团队具有较强的科研和创新能力,近三年,在三大检索系统中共发表论文10多篇。学校特别为生物系制订了人才引进计划,近两年准备引进在生物技术方面有突出贡献的专家、教授2名,985重点院校毕业的博士或在211院校工作过的副教授5名,有计划安排现有的青年教师进入国内外著名的实验室或研究机构进修或学习,以此为基础,构建学院的核心创新团队,组建1个具有一定研究实力的生物制药研究中心,并以此为载体,大力发展本科教育,培养生物工程(以能源生物技术为主要研究方向)和制药工程(以医药生物技术为主要研究方向)本科专业的学生。

1、学科基本情况
原化学与生物工程学院是一个多学科及交叉学科并存的综合性学院,有化学与化学工程、生物技术、环境科学三个一级学科,以及交叉学科覆盖了全院八个专业:化学工程与工艺、应用化学、精细化工、工业分析、商品质量检测、生物制药、制药工程、环境工程,其中已有化学工程与工艺、应用化学两个本科专业。07年3月学校完成了学科专业结构布局调整,进行资源重组,由原来的化学与生物工程学院,新组建成立了化学化工学院和生物系。生物系现有15人,副教授1人,讲师7人,助教4人,全部பைடு நூலகம்有硕士学位,其中博士5人。现有生物制药、制药工程两个专业,2001年以来形成了以能力培养为主线,以基础知识的传授和学习能力的培养、工程观念和创新能力的培养为两个教学重心。全面体现“厚基础、宽口径、重实践、高素质、创造性”整体思路,突出本专业在新药研究与开发方面所形成的特色。

质粒简介
(1)pBR322是研究得最多,使用最早且应用最广泛的大肠杆菌质粒载体之一。

质粒名称pBR322中的“p”代表质粒,“BR”代表两位两位研究者Bolivar和Rogigerus姓氏的字首,“322”是实验编号。

(2)pBR322是一个人工构建的重要质粒,有万能质粒之称。

它是由pSF2124、pMB1及pSC101三个亲本质粒经复杂的重组过程构建而成的。

(3)pBR322具有很多优点,如相对分子质量较小、拷贝数较高等,是最常用的克隆载体之一。

RF3224-CHI 无线接收器
1.0 概述
RF3224-CHI无线接收器,允许 DS7240 控制主机从无线
装置上接收RF无线信号。

2.0 技术指标
• 尺寸(H×W ×D):10.8×15.2 ×3.1(cm)
• 工作环境温度:0°~+65℃
• 频率:315.05MHz
• 电源:一般为 12 VDC,30mA
• 可兼容的控制主机: DS7240V2-EXP,DS7240-EXP
3.0 安装
3.1 安装条件
• 把接收器尽量安装在所有无线探测器的中心部位。

• 天线附近25cm内无影响物。

• 避免把接收器安装在金属表面或带电的线路上。

如无
法避免,安装时天线应高于金属面。

• 避免把接收器安装在可能接触潮湿空气的地方。

• 安装位置越高(天线周围无金属物),接收距离就越长。

3.2 安装墙壁防拆开关(可选)
请按照下面的步骤来启用墙壁防拆开关。

如果不需要墙壁防拆
开关,请看 3.3。

• 卸下外壳
在此处插入螺丝刀
按下外罩定位梢,移去内部塑料罩。

装上内部塑料罩。

如图把弹簧安装于接收器后部防拆开关的轴杆上。

注意:安装弹簧时,请不要将弹簧强迫性地压在轴杆上而导
致轴杆损坏。

3.3 安装接收器
• 确定接收器的安装位置。

• 以探测器底座为模板,在墙壁上标出探测器的安装孔。

• 如图在天线接口处插入天线,并且用螺钉进行固定。

3.3 设置接收器的选项
改变设置的时候,需要先断掉电源后进行设置,然后再上电让新的设置功能生效。

详细的资料,请参照DS7240 控制主机的无线安装指引。

• 合适地设置监察选项。

• 设置合适的接收器地址码选项。

4.0 接线和通电
4.1 将接收器连接到控制主机
• 断掉控制主机的电源。

• 如图将 DS7240 主机上的 Option/SDI 总线的接线端子
和接收器的接线端子连接起来,使用的线径为 0.8mm 或 1.0mm
以上。

接收器和主机之间的距离不要超过 300m 。

总线上增加其他的设备可能会减少这个连线距离。

不推荐使用屏蔽线,也不需双绞线。

• 盖上接收器的外壳。

• 给控制主机上电。

接收器中央的红色 LED 灯会变亮。

4.2 LED 灯的状态
下面的表列出 LED 灯的状态和对应的接收器的状态。

5.0 主机编程
请参照 DS7240-EXP 或 DS7240V2-EXP 控制主机的无线安装指引。
