测定意义:AKP是指在碱性条件下催化蛋白质肽键水解的酶类,属于丝氨酸蛋白酶。

此外,该酶还能够水解酯键、酰胺键,具有转酯及转肽的功能。

该酶是主要工业用酶之一,广泛应用于制药、丝绸、食品、制革等行业。

测定原理:在碱性条件下,AKP水解酪蛋白生成酪氨酸;在碱性条件下,酪氨酸还原磷钼酸生成钨蓝;钨蓝在680nm有特征吸收峰,测定680nm吸光度增加速率,来计算AKP活性。

自备仪器和用品:可见分光光度计/酶标仪、微量玻璃比色皿/96孔板、水浴锅、磁力搅拌器、可调式移液枪、0.5mL EP管和蒸馏水。

试剂组成和配制:试剂一:液体×1 瓶,4℃保存。

试剂二:粉剂×1 瓶,4℃保存。

临用前加 5mL 蒸馏水溶解。

试剂三:粉剂×1 瓶,4℃避光保存。

临用前加入 5mL 试剂一,沸水浴中磁力搅拌溶解。

试剂四:粉剂×1 瓶,4℃保存。

临用前加 20mL 蒸馏水溶解。

试剂五:液体×1 瓶,4℃保存。

标准品:液体×1 支,0.25μmol/mL 标准酪氨酸溶液,4℃保存。

粗酶液提取:1. 组织:按照组织质量(g):试剂一体积(mL)为1:5~10的比例(建议称取约0.1g组织,加入1mL试剂一)冰浴匀浆,8000g,4℃离心10min,取上清,即粗酶液。

2. 血清或培养液:直接测定。

3. 细菌、真菌:按照细胞数量(104个):试剂一体积(mL)为500~1000:1的比例(建议500万细胞加入1mL试剂一),冰浴超声波破碎细胞(功率300w,超声3秒,间隔7秒,总时间3min);然后8000g,4℃,离心10min,取上清置于冰上待测。

测定操作:1. 分光光度计/酶标仪预热30min,调节波长到680nm,蒸馏水调零。

由于编辑部其他人都出差在外,小编夜雨有幸成为了这款键盘的第一个体验者。

它是否真的如传说中的那么犀利?让小编为你一一道来。

全方位的位置调节看到键盘包装盒上的说明,小编就感到压力很大:除了英文法文和德文,一句中文都没有看到!看来国家普及英语四六级的确是有道理的,否则连个键盘说明都看不懂就悲剧了。

好在小编当年也做过计算机外文图书的编辑,囫囵吞枣地看说明书加上手动摸索,总算是把这个键盘的使用方法弄了个八九不离十。

入手键盘就发现,这个键盘“长”的很特别。

首先是在普通的104键键盘的两头,分别加上了6个功能按键;其次是键盘的最上部分,有一个发光显示屏,上面有一系列符号;最后就是键盘线,它有两个USB、一个麦克风以及一个耳机总共4个插头!4个插头的数据线并不是单独的,它们被收束在一根直径达0.6mm的粗线当中。

从外形看,最特别的无疑是本键盘附带的一个手托构件了。

这个可拆卸的手托与键盘主体一样,采用了银黑配色设计。

由塑料磨砂材质制造。

这个手托不仅可以随意拆卸。

还能够通过调节一个内置的卡扣来调整手托与键盘的相对位置。

当小编把手托安装上去并找到一个合适的位置之后,发现双手搁在上面打字的确变得轻松了不少,时间一长感觉更为明显。

The ropes should be pulled down in the locks so that they are sufficiently fastened to avoid slipping of the ropes when lifting.If the patient is large and heavy, it may be neces-sary to pull the ropes from the outside in on the V-shaped suspension in order to increase the width between the connection pointsB7. Designedfor optimum performance in any type of project, the equipment is built according to certainfundamental principles.All basic components are bolted to each other, which enables fast and easy mounting,assembly and servicing. Since the stem is bolted to the base-head, different sizes of stemcan be fitted to suit different sizes of pilot hole. The saddles, too, are bolted to the head.They are therefore easy to re-position, which gives maximum flexibility.The Sandvik system is based on standard components, which contributes greatly to itsflexibility and high availability. All heads have a flat cutting profile which rotates smoothlyand with optimum thrust utilisation.Sandvik reaming heads are easy to adapt to different rock conditions by re-arrangingthe cutters. The effect is to change the spacing between the rows of cemented-carbide but-tons that break the rock. To enable row spacing to be varied, only two types of cutter areneeded on any one reaming head. Placing one or the other type of cutter in differentpositions on the head gives either wide or narrow button-row spacing. The uniquedesign of Sandvik cemented-carbide buttons ensures long service life and the highestpossible rate of penetration.A special gauge saddle system has been developed with the option to reduce the reamerdiameter in two steps simply by changing to diffrent types of saddle. It is drilled using a roller bitwith sealed bearings, together with hollow drill pipes 1.5 m in length with an internationalstandard thread for high–torque applications.The cuttings are removed from the pilot hole with the aid of water flushing. Introducedthrough the center of the drill string, the water flows out of the drill bit and up throughthe annulus between the drill pipes and the hole wall. If required, the pilot drilling can becontrolled by using a directional drilling system.Raise boringWhen the pilot hole breaks through into the lower level, the roller bit is removed and replacedwith a reaming head. The reamer is rotated and pulled back toward the drilling unit. Thecuttings fall by gravity into the chamber at the bottom of the hole, where they aremucked out using a LHD-type loader.Raises up to 6 m in diameter and up to 1000 m in length are not uncommon. The raise-boringmethod is used to produce ventilation shafts, ore passes, manways, penstocks etc.Blind boringWhen a raise is required but there is no access to the upper level, it has to be bored blindfrom below, usually without a pre-drilled pilot hole. A special type of head is required forblind boring. It drills the pilot hole and reams out the raise at the same time. The head isrotated and pushed upward. The cuttings fall out of the hole by gravity.Normal blind raise diameters are from 0.6 to 1.8 m. Since the drill string is undercompression during blind boring, special large-diameter stabilizers are needed to supportthe drill string. The blind-boring method is used to produce so-called slot raises, ore passesand manways.Horizontal boringHorizontal boring is an excellent method in urban construction projects where drilling andblasting is restricted or forbidden and tunnel boring machines (TBMs) are too bulky. First,a horizontal pilot hole is drilled, with the aid of a directional drilling system if necessary.When the pilot bit breaks through, it is removed and replaced with a reaming head. Becausethe hole is horizontal, the reamer must be equipped with a special cuttings removal system.Typical diameters for horizontal reaming are from 0.6 to 4.5 m. The method is used todrill tunnels for cables, escape routes, sewage etc without disturbing the environment unduly.Horizontal boring requires good rock stability.Down boring with a pre-drilled pilot holeIn mines, large fill-holes between 0.6 and 1.8 m in diameter can be bored into stopes usingreaming equipment, provided that a pilot hole can be pre-drilled into the stope. The pilotbit and drill string are then removed and a reamer fitted. The reamer is pushed and rotateddownward, guided by a nosepiece that follows the pilot hole.The cuttings fall by gravity down through the pilot hole. The segmented design comprises abase-head and two removable segments. This enables the reamer to be transported to thecollaring site via small cages and narrow drifts. 【ASTK/STK牌电机主要分类】:感应电机,感应调速电机,可逆电机,可逆调速电机,电磁刹车电机,调速附电磁刹车电机,力矩电机,离合刹车电机,调速附离合刹车电机。

【感应电机】:适用于单方向长时间的连续运行。

与电容器连接后只需连接交流电源,就可轻易运行。

【单相力矩电机】:适用于卷取作业,如收料,放料,有堵转功能。

通过改变对电动机的施加压电压,可调节转矩:力矩电动机有大起动转矩与下垂特性。

电动机的转矩与施加电压的2次方成正比变化,因此,负载与施加电压改变时,则转速也发生变化。

卷取作业:对输出物体连续进行一定速度固定张力卷取时,卷框直径加倍,则转矩也变为2倍,速度则减半。

因此,可广泛用于卷取用途。

齿轴电机力矩电机与减速机组合使用,可以实现减速至目标速度或者提高转矩。

功率:3W-60W,电压单相110V/220V。

【电磁刹车电机】:以电源控制刹车动作。

原理/结构:1:内装失电电磁制动器。

2:电源断电,制动器衔铁释放同时弹簧使衔铁复位压紧摩擦盘,电机轴被摩擦盘抱住,制动保持负载。

特点:1:电源断电即制动,保持负载。

2:过转量约2-3转。

3:单相、三相齐全。

【蜗轮减速机】:蜗轮减速电机具有一定的自製动性能,出力轴与电动机成90度转向,具有精简,节省安装空间特性.本系列产品已经把减速器与电动机配置在一起使用方便,可用于机械设备、升降机构、输送机构…等。

可搭配电机功率:40W,60W,90W,120W【调速器分类】:组合型调速器,分离型调速器,调速附电子刹车器,电子刹车器,力矩电机控制器。

【US组合型调速器】: 外露型单相AC电机调速器,调整调速器面板上的旋钮,即可控制电机运转速度的快慢。

内装启动电容。

透过连接线直接与电机连接,免去烦琐的接线,与马达连接后通电即可,在面板上即可作控制, 无法瞬时正逆转/停止。

【电子式刹车器】:刹车器能于0.1秒以内使马达瞬间停止(空载时)。
