第43 卷第 1 期2024 年1 月Vol.43 No.119~31分析测试学报FENXI CESHI XUEBAO(Journal of Instrumental Analysis)生物光学成像技术在组织穿透性方面的研究进展张玉敏,王富,林俐*,叶坚*(上海交通大学生物医学工程学院,上海200030)摘要:光学成像因灵敏度高、特异性强、无电离辐射、低成本、丰富的候选探针、可获取细胞/分子水平信息和可实时检测等优势,在临床前的基础研究和临床诊断与治疗领域具有巨大的应用价值。
但由于生物组织对光子的高散射与高吸收特性,光学成像的组织穿透深度通常非常有限,很大程度上限制了其在深部病变活体生物医学检测方面的应用,研究者们对此做了大量的努力。
随着科学技术的发展,光学技术的组织检测深度已覆盖微米到厘米甚至分米以上的范围,在生物检测、成像、诊断、术中导航等领域展现出了广阔的应用前景。
该文从常见的光学成像技术入手,对荧光成像、生物/化学发光成像、光声成像以及拉曼成像在组织穿透性方面的研究进展进行了总结与讨论,并对这些光学成像技术未来在组织穿透方面的主要研究方向进行了展望。
关键词:生物光学成像;组织穿透性;深穿透拉曼光谱中图分类号:O657.3;R318文献标识码:A 文章编号:1004-4957(2024)01-0019-13Advances in Tissue Penetration by Optical Imaging TechniquesZHANG Yu-min,WANG Fu,LIN Li*,YE Jian*(School of Biomedical Engineering,Shanghai Jiao Tong University,Shanghai 200030,China)Abstract:Optical imaging has great potential for application in the field of preclinical basic research and clinical diagnostics and therapeutics,due to its advantages of high sensitivity and specificity,non-ionizing radiation,simplicity of equipment,low cost,rich nanoprobe candidates,ability to ob⁃tain cellular/molecular level information and real-time acquisition capability.However,due to the nature of high scattering and absorption of photons in biological tissues,optical imaging is usually limited by the shallow tissue penetration depth,which largely limits its usage for in vivo biomedical detection of deep-seated lesions. A lot of efforts have been done by researchers to overcome this is⁃sue.This paper summarizes and discusses the progress of various common optical imaging tech⁃niques,such as fluorescence imaging,bioluminescence/chemiluminescence imaging,photoacous⁃tic imaging,and Raman imaging,in terms of their research progress in tissue penetration. With the development of science and technology,the tissue detection depths of optical modalities have cov⁃ered a range from microns to centimeters or even to decimeters,and have shown broad application prospects in the fields of biological detection,imaging,diagnosis,intraoperative navigation,and so on. 结构光照明超分辨光学显微成像技术与展望
摘要:结构光照明显微镜(StructuredIlluminationMicroscopy,SIM)通过结构化照明在频率域以空间混频的方式将物体高 频信息载入光学系统的探测通带内实现突破衍射极限的 超 分 辨 光 学 显 微 成 像。SIM 凭 借 其 较 低 的 激 发 光 强、对 荧 光 染 料的非特异性需求以及快速的宽场成像优势已成为活细胞超分辨光学显微成像方面应用最多的技术。本文系统回顾了 SIM的技术进展,对 SIM的基本原理与实现方法进了详细的分析,重点介绍了本课题组研发的基于光谱分辨的单光子激 发超分辨显微镜和结合自适应光学的双光子激发超分辨显微镜这两种最新的 SIM技术,最后简要讨论了 SIM技术在生 物成像中的应用及未来发展方向。 关 键 词:超分辨成像;结构光照明显微镜;光学传递函数 中图分类号:O436;O438.2 文献标识码:A doi:10.3788/CO.20181103.0307

偏振法对活体兔血糖浓度预测的平均绝对相 对差 (MeanAbsoluteRelativeDifference,MARD) 已低至 1166mg/dL[30],可见其是一种极具潜力 的无创血糖检测方法。未来应从优化测量系统、 改进测量原理以及引进校正算法等方面减少甚至 消除人眼运动引起的双折射对测量精度的影响。 此外,偏振法一般选择眼前房作为测量部位,故还 需考虑眼前房水中糖浓度与血糖浓度的延迟时间 以及人眼检测的安全性问题。延迟时间的存在会 导致测得的血糖浓度并非真正意义上的实时数 据,这对于需要严格控制血糖浓度的糖尿病患者 或重症监护组(IntensiveCareUnit,ICU)患者是极 其不利的。 2.2 光学相干断层成像法

一、出版情况
《生物医学光学― 原理和成像》(Biomedical Optics ― Principles and Imaging)一书由John Wiley & Sons出版集团出版,全书362页,南开大学图书馆馆藏版本为2007年第一版。
在两位作者中,Lihong V. Wang教授任职于圣路易斯华盛顿大学生物医学工程系,而Hsin-I Wu教授任职于得克萨斯农机大学生物医学工程系。
二、内容简介
《生物医学光学― 原理和成像》是生物医学光子学领域内为数不多的教科书中的一种,是作者在使用了近十年的研究生讲义的基础上改编而成的。
其主要特点是在介绍基本概念的同时,给出了严格理论数学推导和数值计算方法,并有针对性地提供了大量课后习题。
其另一个特点是把介绍基本原理和讲解实际的科学研究有机结合起来,每章后面都提供了丰富的与相应研究有关的参考文献。
由于作者之一Wang Lihong教授为生物医学光子学领域内资深学者,现任国际生物医学光学学会的主席,并在本书涵盖的多个研究领域均有建树,所以本书一经出版就引起了广泛的关注。
本书内容可分为两个大部分。
第一部分包括第1到第7章,系统介绍了光与生物组织的相互作用以及光在生物组织内的传输机理,重点为第3章的描述光在生物组织内传输过程的蒙特卡罗模拟。
第二部分包括第8章到第13章,分别描述了当前各种流行的生物医学成像方法。
由于本书触及到了生物医学光子学的大部分研究方向,是比较理想的研究生入门教材,由于理论性较强,需要学生具有一定的物理和数学基础。
除作为教材之外,由于本书内容详实文献全面,也可作为研究工作者了解相关领域、掌握最新发展动态的参考读物。
