【Abstract】Objective:To study the expression and clinical significance of claudin－6and clusterin protein in gas-

tric carcinoma tissues and cell lines．Methods:The expression of claudin－6and clusterin protein were detected by

immunohistochemistry and immunocytochemistry method in normal gastric tissue，gastric dysplasia，gastric carcinoma

and gastric carcinoma cell lines(MKN28，SGC7901，BGC823)．Results:The positive expression rates of claudin－6

and clusterin in gastric carcinoma tissues，gastric hyperplasia and normal gastric mucosa were53．3%，65%，100%

and60%，75%，100%，and differences among the3groups were remarkly significant(P＜0．05，P＜0．05)．The ex-

pression of claudin－6and clusterin protein were related to tumor depth of gastric carcinoma and lymph nodes metas-

tasis，but they were not correlated with patient＇s age，sex and vessel invasion．In addition，the expression of claudin－6

protein was positively related to that of clusterin protein(r=0．899，P＜0．05)．In gastric carcinoma cell lines claudin

－6and clusterin were mostly expressed in cytoplasm and cell membrane．On the basis of the degree of differentia-

tion，the immunostaining intensities of clusterin among3cell lines(MKN28，SGC7901，BGC823)were reduced，but

those of claudin－6were not remarkly significant．Conclusion:Loss of the expression of claudin－6and clusterin play

an important role in the carcinogenesis and subsequent progression of gastric carcinoma and might be used as mean-

ingful markers of judging biological behavior of gastric carcinoma．

【摘要】目的：探讨claudin－6及clusterin在胃癌组织及胃癌细胞中的表达及其临床意义。方法：应用免疫

组织化学及免疫细胞化学法检测claudin－6及clusterin在正常胃黏膜、非典型增生胃黏膜、胃癌组织及胃癌

细胞株（MKN28、SGC7901、BGC823）中的表达。结果：胃癌组织、非典型增生胃黏膜及正常胃黏膜中claudin－

6及clusterin的阳性表达率分别是53．3%、65%、100%及60%、75%、100%，二者的阳性表达率在三组间有统

计学差异（均P＜0．05）。在不同浸润深度及有无淋巴结转移组内claudin－6及clusterin阳性表达的差异性

有统计学意义（P＜0．05），而在不同性别、年龄及脉管侵犯组内无统计学差异（P＞0．05），Spearman等级相关

性分析显示claudin－6与clusterin表达呈正相关（r=0．899，P＜0．05）。在三株胃癌细胞中claudin－6与

clusterin均阳性表达在细胞膜和细胞质中。依分化程度clusterin在MKN28、SGC7901、BGC823三种细胞系中

免疫染色强度逐步减弱，claudin－6在三种细胞系中免疫染色强度无明显差别。结论：claudin－6与clusterin

的缺失与胃癌发生发展及浸润转移有关，有望成为反映胃癌生物学行为有价值的标志物。

有研究表明多种上皮组织来源的肿瘤claudins表达异常，并与这些肿瘤的发生、发展密切相关［1］。胃癌是常见的消化道恶性肿瘤，有关其发生、发展的机制尚未完全阐明。为此，我们采用免疫组化法检测claudin－6蛋白在正常胃黏膜、非典型增生及胃癌组织中的表达及其与clusterin表达的关系，以探讨其在胃癌的发生、发展及浸润转移中的作用。

1资料与方法

1．1资料

全部病例来自邢台市人民医院病理科2008年2月－2009年11月存档石蜡包埋标本。胃癌组45例，包括男性33
