Inhibitors, Agonists, Screening Libraries Data SheetBIOLOGICAL ACTIVITY:TAK–438 (free base) is a novel P–CAB (potassium–competitive acid blocker) that reversibly inhibits H+/K+ ATPase with IC50 of 19 nM (pH 6.5), controls gastric acid secretion.IC50 value: 19 nM [1]Target: H+/K+ ATPasein vitro: TAK–438 is a pyrrole derivative with a chemical structure that is completely different from the P–CABs developed to date.TAK–438 inhibits gastric H+/K+ ATPase activity in a concentration–dependent manner. Under neutral conditions (pH 7.5), theinhibitory activity of TAK–438 is almost the same as that under weakly acidic conditions (pH 6.5). TAK–438 does not inhibit Na+/K+ATPase activity even at concentration 500 times higher than their IC50 values against gastric H+/K+ ATPase activity. TAK–438 inhibits gastric H+/K+ ATPase in a K+ competitive manner with Ki of 3 nM [2].in vivo: TAK–438 inhibits basal gastric acid secretion in a dose–dependent manner, and the ID50 value is 1.26 mg/kg. Intravenous administration of TAK–438 dose–dependently increases the pH of the gastric perfusate, and the increase in pH is sustained for 5 h after administration. At the 1 mg/kg dose, the pH plateaues 90 min after administration, and the highest pH value reached is 5.9 [2]. In addition, TAK–438 shows a potent and longer–lasting inhibitory effect on the histamine–stimulated gastric acid secretion in rats and dogs. TAK–438 shows significant antisecretory activity through high accumulation and slow clearance from the gastric tissue. TAK–438is unaffected by the gastric secretory state, unlike PPIs [3].PROTOCOL (Extracted from published papers and Only for reference)Enzyme assay [2]A 2–μl volume of TAK–438 solution was preincubated with 12 μl of 0.002% HCl solution for 5 min and incubated with 4 μl (0.5 mg/ml)of pig gastric vesicles for 5 min at room temperature. After neutralizing the enzyme–compound mixture with 2 μl of 500 mM HEPES–Tris buffer (pH 6.5), a 1.1–μl volume of the mixture was diluted with 39 μl of 50 mM HEPES–Tris buffer containing 12.5 μMvalinomycin and 5 mM MgCl2. As an undiluted control, 4.9 μl of TAK–438 was added to dilution mixtures at the same concentration as that of the original incubation; for the dilution treatment group, 4.9 μl of vehicle was added to the dilution mixtures. After 60 min, the reaction was initiated by adding 5 μl of 2 mM ATP with or without 10 mM KCl, and the reaction mixture was incubated for 60 min at 37°C. The reaction was then stopped, and the ATPase activity was determined as described previously.Animal administration [2]Drugs and the vehicle were given orally (2 ml/kg) to rats in a blind manner. The pylorus was ligated under light anesthesia 1 h after drug and vehicle administration, and the abdomen was closed by suturing. At 3 h after the pyloric ligation, the rats were sacrificed by CO2 asphyxiation, and their stomachs were removed. The gastric contents were collected and centrifuged at 3000 rpm for 10 min.
