实验原理:SDS-PAGE是对蛋白质进行量化,比较及特性鉴定的一种经济、快速、而且可重复的方法。
该法是依据混合蛋白的分子量不同来进行分离的。
SDS是一种去垢剂,可与蛋白质的疏水部分相结合,破坏其折叠结构,并使其广泛存在于一个广泛均一的溶液中。
SDS蛋白质复合物的长度与其分子量成正比。
在样品介质和凝胶中加入强还原剂和去污剂后,电荷因素可被忽略。
蛋白亚基的迁移率取决于亚基分子量。
试剂和器材:试剂:1. 5x样品缓冲液(10ml): 1mol/L的Tris-HCl,5ml 50%甘油,2ml 10%的SDS,巯基乙醇,1ml 1%溴酚蓝,蒸馏水。
可在4℃保存数周,或在-20℃保存数月。
2. 凝胶贮液:在通风橱中,称取丙烯酰胺30g,甲叉双丙烯酰胺,加重蒸水溶解后,定容到100ml。
过滤后置棕色瓶中,4℃保存,一般可放置1个月。
3. 分离胶缓冲液: Tris ,加1mol/L HCl 48ml,加重蒸水80ml 使其溶解,调,定容至100ml,4℃保存。
4. 浓缩胶缓冲液: Tris ,加1mol/L HCl 48ml,加重蒸水80ml 使其溶解,调,定容至100ml,4℃保存。
5. TEMED(四乙基乙二胺)原液%过硫酸铵(用重蒸水新鲜配制)7. Tris-甘氨酸电极缓冲液:称取Tris ,甘氨酸,加蒸馏水约900ml,调后,用蒸馏水定容至1000ml。
置4℃保存,临用前稀释10倍。
8. 考马斯亮蓝G250染色液:称100mg考马斯亮蓝G250,溶于200ml蒸馏水中,慢慢加入 70%的过氯酸,最后补足水到250ml,搅拌1小时,小孔滤纸过滤。
器材:电泳仪,电泳槽,水浴锅,摇床。
实验操作(一)样品制备将蛋白质样品与5X样品缓冲液(20ul+5ul)在一个Eppendorf 管中混合。
放入100℃加热 5-10min,取上清点样。
(二)分离胶及浓缩胶的制备1 将玻璃板、样品梳、Spacer用洗涤剂洗净,用ddH2O 冲洗数次,再用乙醇擦拭,晾干;2 将两块洗净的玻璃板之间加入Spacer,按照Bio-Rad Mini Ⅱ/Ⅲ说明书提示装好玻璃板;3 按如下体积配制10%分离胶 ml,混匀;ddH2O mlmol/LTris-HCl pH= ml30% Acr-Bis ml10% SDS 80 ul10%AP 56 ulTEMED 6 ul4 向玻璃板间灌制分离胶,立即覆一层重蒸水,大约20 min后胶即可聚合;5 按如下体积配制6%浓缩胶 ml,混匀;ddH2O mol/LTris-HCl pH= 30% Acr-Bisml 400 ul 600 ul10% SDS 10% AP TEMED36ul 24ul 4ul6 将上层重蒸水倾去,滤纸吸干,灌制浓缩胶,插入样品梳;7 装好电泳系统,加入电极缓冲液,上样20 μl;8 稳压200V,溴酚蓝刚跑出分离胶时,停止电泳,约需45 min~1hr.9 卸下胶板,剥离胶放入染色液中,室温染色1~2 hr;加入脱色液,置于80 rpm脱色摇床上,每20 min更换一次脱色液(10 ml冰乙酸;45 ml乙醇;45 ml蒸馏水)至完全脱净。

5. 程序:5.1. 定义:SDS-聚丙烯酰胺凝胶电泳测定蛋白分子量,是根据大多数蛋白都能与阴离子表面活性剂十二烷基硫酸钠(SDS)按重量比结合成复合物,使蛋白分子所带的负电荷远远超过天然蛋白分子的净电荷,消除了不同蛋白分子的电荷效应,使蛋白分子相对迁移率(Rf)的大小完全取决于分子量的高低,因此可从已知分子量的标准蛋白的对数和相对迁移率所作的标准曲线中求出供试品的分子量。

5.4. 溶液的配制:5.4.1.丙烯酰胺—N,N′—亚甲基双丙烯酰胺贮备溶液:称取丙烯酰胺29.2g。
N、N′—亚甲基双丙烯酰胺0.8g,加水溶解,并稀释至100ml,过滤至棕色瓶中,4℃保存。
5.4.2.三羟甲基氨基甲烷缓冲液(pH8.8):称取三羟甲基氨基甲烷18.15g,加水溶解,用2mol/L盐酸溶液调节pH值至8.8,加水稀释至100ml,4℃保存。
5.4.3.三羟甲基氨基甲烷缓冲液(pH6.8):量取上述三羟甲基氨基甲烷缓冲液(pH8.8)适量,加水(1:3)稀释,用2mol/L盐酸液调节pH值至6.8,4℃保存。
5.4.4.10%十二烷基硫酸钠溶液:称取十二烷基硫酸钠10g,加水溶解,并稀释至100ml,室温保存。
5.4.5.供试品缓冲液:水 4.8ml三羟甲基氨基甲烷缓冲液(6.8) 1.2ml甘油 1.0ml10%SDS溶液 2.0ml0.5%(W/V)溴酚蓝溶液0.5mlβ-巯基乙醇0.5ml总体积10.0ml5.4.6.电极贮存液(pH8.3):称取三羟甲基氨基甲烷15g,甘氨酸72g,SDS5g,加水溶解,并稀释至1000ml,4℃保存。
SDS-聚丙烯酰胺凝胶电泳
聚丙烯酰胺凝胶电泳PAGE作用:用于蛋白质与寡糖核苷酸的分离。
作用原理聚丙烯酰胺凝胶电泳是网状结构,具有分子筛效应,它具有两种形式,一种是非变性聚丙烯酰胺凝胶,蛋白质在电泳中保持完整的状态,蛋白在其中依三种因素分开:蛋白大小,形状和电荷。
而SDS-PAGE仅根据蛋白分子量亚基的不同而分离蛋白。
这个技术首先是196 7年由shapiro建立,他们发现在样品介质和丙烯酰胺凝胶中加入离子去污剂和强还原剂后,蛋白质亚基的电泳迁移率主要取决于亚基分子量的大小,电荷因素可以忽视。
作用:SDS是阴离子去污剂,作为变性剂和助溶试剂,它能断裂分子内和分子间的氢键,使分子去折叠,破坏蛋白分子的二、三级结构。
而强还原剂如巯基乙醇,二硫苏糖醇能使绊胱氨酸残基间的二硫键断裂。
在样品和凝胶中加入还原剂和SDS后,分子被解聚成多肽链,解聚后的氨基酸侧链和SDS结合成蛋白- SDS胶束,所带的负电荷大大超过了蛋白原有的电荷量,这样就消除了不同分子间的电荷差异和结构差异。
SDS-PAGE一般采用的是不连续缓冲系统,于连续缓冲系统相比,能够有较高的分辨率。
浓缩胶的作用是有堆积作用,凝胶浓度较小,孔径较大,把较稀的样品加在浓缩胶上,经过大孔径凝胶的迁移作用而被浓缩至一个狭窄的区带。
当样品液和浓缩胶选TRIS/HCL缓冲液,电极液选TRIS/甘氨酸。
电泳开始后,HCL解离成氯离子,甘氨酸解离出少量的甘氨酸根离子。
蛋白质带负电荷,因此一起向正极移动,其中氯离子最快,甘氨酸根离子最慢,蛋白居中。
电泳开始时氯离子泳动率最大,超过蛋白,因此在后面形成低电导区,而电场强度与低电导区成反比,因而产生较高的电场强度,使蛋白和甘氨酸根离子迅速移动,形成以稳定的界面,使蛋白聚集在移动界面附近,浓缩成一中间层。
此鉴定方法中,蛋白质的迁移率主要取决于它的相对分子质量,而与所带电荷和分子形状无关。
补充信息聚丙烯酰胺凝胶电泳简称为PAGE,是以聚丙烯酰胺凝胶作为支持介质的一种常用电泳技术。
SDS聚丙烯凝胶电泳实验方案
SDS聚丙烯凝胶电泳一、试剂(1)分离胶缓冲液(Tris-HCl缓冲液PH8.9):取1mol/L盐酸48mL,Tris 36.3g,用无离子水溶解后定容至100mL。
(2)浓缩胶缓冲液(Tris-HCl缓冲液PH6.7):取1mol/L盐酸48mL, Tris 5.98g,用无离子水溶解后定容至100mL。
(3)30%分离胶贮液:配制方法与连续体系相同,称丙烯酰胺(Acr)30g及N,N’-甲叉双丙烯酰胺(Bis)0.8g,溶于重蒸水中,最后定容至100ml,过滤后置棕色试剂瓶中,4℃保存。
(4)10%SDS溶液:SDS在低温易析出结晶,用前微热,使其完全溶解。
(5)1%TEMED;(6)10%过硫酸铵(AP):现用现配。
(7)电泳缓冲液(Tris-甘氨酸缓冲液PH8.3):称取Tris 6.0g, 甘氨酸28.8g, SDS1.0g, 用无离子水溶解后定容至1L。
(8)样品溶解液:取SDS 100mg,巯基乙醇0.1mL,甘油1mL,溴酚蓝2mg,0.2mol/L,pH7.2磷酸缓冲液0.5mL,加重蒸水至10mL(遇液体样品浓度增加一倍配制)。
用来溶解标准蛋白质及待测固体。
(9)染色液:0.25g考马斯亮蓝G-250,加入454mL 50%甲醇溶液和46mL冰乙酸即可。
(10)脱色液:75mL冰乙酸,875mL重蒸水与50mL甲醇混匀。
二.实验步骤1、组装模具1)用海绵或纱布沾取少量清洁剂(也可用纯酒精浸泡),擦洗干净两块玻璃板,自来水冲洗3-5遍,然后,再用无水乙醇冲洗2遍,晾干待用。
2)将已冲洗好的玻璃板心均匀涂上凡士林,将玻璃板心放在玻璃板的两侧(下面一定要靠边),再将另一块玻璃板盖上待用。
3)将隔条安置好,再将事先准备好的琼脂溶液倒入电泳仪的槽中,再将两块玻璃板插入槽中,有缺口的放在里侧,用夹子夹好待用。
2、制胶1)20ML10%分离胶/ML分离胶贮液 6.66分离胶缓冲液(PH8.9) 2.510%SDS 0.21%TEMET 2.00重蒸水8.5410%AP 0.1(AP最后加入,注意观察是否漏胶)2)水封,等待20-30分钟,用滤纸吸干3)10ML 3%浓缩胶/ML分离胶贮液 3.0浓缩胶缓冲液(6.8) 1.2510%SDS 0.11%TEMED 2.00重蒸水 4.610%AP 0.14)插上齿梳,确保无气泡。
