Oligo DNA的Tm值 引物设计软件都可以给出Tm,与引物长度,碱基组成, 引物使用缓冲的离子强度也有关。长度为25mer以下的引物, Tm计算公式为: Tm = 4℃(G + C)+ 2℃(A + T) 引物Tm值与PCR产物Tm值相差一般不超过30℃,一般采 用较引物Tm值低5℃作为PCR退火温度。兼并碱基Tm值可 以折算。 酶切位点和保护碱基不计算在内,而只计算与模板互补的 碱基序列的Tm。

根癌农杆菌(Ti质粒)
Ti质粒是约200-500kb的环状DNA分子, Ti质粒具有五个主要功能区域,它们分别是 T-DNA、质粒转移(plasmid transfer)、冠瘿 碱代谢(opine catabolism)、复制原点(ori)和 毒性区域(vir)。 T-DNA中直接参与转移并整合到植物染色 体上的序列,仅是T-DNA两端与其它序列交 界处的25bp不完全直接重复(imperfect direct repeats),右端的序列称为右界(right border, RB), 左端的为左界(left border,LB)。 T-DNA区域内的所有基因与转移无关,所以 将致瘤基因全部缺失即卸甲(disarmed)后, 将其它序列插入到这个区域,形成的T-DNA 仍可将RB至LB内的序列转移并整合到植物 基因组。 Vir区的毒性基因是T-DNA转移所必需的。

第六章:以大肠杆菌为宿主的克隆载体
• 与pBR322相比,pBR325有两个优点: 1)多了一个用于克隆的限制性识别位点(pBR322也有一
个EcoRI切点,但它既不在ampR,也不在tetR基因里) 2)第三个用作选择标记的抗生素抗性基因。

• pAT153和pBR327都是多拷贝质粒 • 这两种质粒均来自pBR322,在构建时,并非关系
pGEM3Z载体也含有ampR 和lacZ基因,并且在lacZ基因内 部有一个限制性内切酶位点,此外pGEM3Z载体中限制性 内切酶位点两边还多了两个短片段,每一个片段都可充当 启动子,可附着RNA聚合酶进行转录。外源DNA在限制 性内切酶位点被导入pGEM3Z载体后,重组后的pGEM3Z 质粒和提纯的RNA聚合酶在试管中可实现RNA的转录。 pGEM3Z质粒携带的启动子并不是被大肠杆菌RNA聚合酶 识别的标准启动子,而是被T7噬菌体编码的RNA聚合酶 及SP6噬菌体编码的RNA聚合酶专一识别,因此体外RNA 合成系统选择的是病毒RNA聚合酶。

本研究以pCAMBIA1300载体为基础,基因操作去除pCAMBIA1300质粒上的潮霉素抗性基因和花椰菜花叶病毒的35S启动子序列,构建了两种只含有水稻蜡质基因启动子引导蜡质基因反义片段的表达载体,p13AWY-1和p13AWY-2。

其中p13AWY-1表达载体含有一个由水稻蜡质基因启动子、第一内含子、反义蜡质基因(Waxy pro+intron1+anti-Waxy)的融合基因单元;而p13AWY-2表达载体含有两个正向排列的Waxy pro+intron1+anti-Waxy融合基因单元。

我们通过构建水稻反义蜡质基因安全性表达载体,试图为植物安全性表达载体的构建提供一种思路,为今后大规模商业化采用转基因技术改良农作物遗传特性提供安全的转基因方法。

The construct uses E.coli gusA with an intron (from the castor bean catalase gene) inside the coding sequence to ensure that expression of glucuronidase activity is derived from eukaryotic cells, not from expression by residual A.tumefaciens cells. This vector is suitable for insertion of other genes of interest containing their own promoter and terminator. Researchers can excise the gusA gene and insert their own gene of interest in its place or use these vectors to create fusions of gusA with their gene of interest.
