节段性骨缺损修复过程中几种微量元素含量的研究

摘要:目的探讨节段性骨缺损修复过程中,微量元素钙、锌、镁、铜含量变化的规律。方法取7只雄性新西兰大 白兔尺桡骨制成长1.5 cm脱钙骨基质、深低温冷冻骨、深低温冷冻脱钙骨基质84枚,之后进行不同的构建后分别移 植于40只同种大白兔双侧桡骨相同长度骨缺损处(随机分为四组,80肢共8组,其中10肢不移植任何材料作为空白 对照组),于术后l、2、4、8、12周各组分别杀死2只。于移植区取材进行X线、组织学切片、微量元素测定等检查,并对 结果行统计学分析。结果a)除空白对照组骨缺损未愈合外,其余各组均骨性愈合ob)骨缺损修复后第2周末。缺损区 钙、镁、锌达峰值,到第4周末钙、镁降至谷底,而锌仍维持峰值。然后三者共同上升,至第8周末均达第2个峰值。之后 钙、镁轻度下降,而锌轻度卜升。e)铜在骨缺损修复的头4周内一直稳定在低水平,之后开始轻度上升,8周末开始明 显I二升,至第12周末达其峰值。结论a)钙作为评价成骨活性大小的指标,取材应有时间限制;b)骨锌、镁值町能是有 用的评价骨缺损修复材料成骨活性的指标;它们值低,成骨活性高,值高则成骨活性低;c)骨铜值不宜作为评价骨缺损 修复材料成骨活性的指标。

长期以来,在动物实验中对骨折愈合的评价往往主要依 靠X线片和骨痂标本的组织学及组织化学观察[1 J。然而。对 骨痂愈合的组织学评分方法本身具有很大的主观性,H得到 的是统计学上的计数资料,带有一定的局限性,因此需要寻 找能定量反映骨折愈合的较为理想、客观的计量指标心]。有 作者实验表明[3],骨痂灰度,骨矿物质含量以及骨的生物力 学参数之间有着明湿的相关关系,可以对骨折愈合进行综合 评价。本实验对节段性骨缺损修复过程中骨矿的几种代表微 量元素钙(Ca)、锌(Zn)、镁(Mg)、铜(Cu)的含量变化作r初 步研究,对它们是否可以作为骨折愈合的评价指标提出了初 步见解。 1材料和方法 1.1兔骨缺损修复材料的制作7只新西兰大白兔(雄性, 2~2.5 kg,济南农科院畜牧所种兔厂),取长1.5 ClTI尺桡骨 共84枚,12枚经一80C深低温冰箱冷冻、脱水、脱脂、干 燥,60CO照射等制成深低温冷冻骨;另12枚经同样卜述程 序处理后再在0.6 N盐酸中脱钙等制成深低温冷冻脱钙骨 基质;另60枚制成脱钙骨基质。 1.2’骨缺损支架材料的构建A1:指上述所制保持完整管 状的脱钙骨基质;A::指A.一劈为二,1/2旋转180。与另一 半组合;B。:指A,复合碱性成纤维细胞生长因子1 000 IU (将含1 000 IU碱性成纤维细胞生长因子的水溶液0.1 mL 滴人材料髓腔2 h);B。:指A:复合碱性成纤维细胞生长因子 1 000 IU;C。:指髓腔中注满宿主兔的自体骨髓的A。;C。:空 白对照组,指实验兔骨缺损处不植入任何材料;D,:指上述所 制的保持完整管状的深低温冷冻脱钙骨基质;D::指上述所 制的保持完整管状的深低温冷冻骨。 1.3 骨缺损动物模型制作、修复材料植入及动物实验设计
