The p ET-­‐51b(+) v ector c arries a n N-­‐terminal S trep•Tag® I I c oding s equence (1) f ollowed by a recognition site for enterokinase, and an optional C-­‐terminal His•Tag® coding sequence. The presence of two "gentle elution" tags at both the N-­‐terminus and C-­‐terminus is ideal for dual purification strategies designed to isolate full-­‐length fusion proteins (2). U nique r estriction s ites a re s hown o n t he c ircle m ap. N ote t hat t he s equence is numbered by the pBR322 convention, so the T7 expression region is reversed on the circle map. The cloning/expression region of the coding strand transcribed by T7 RNA polymerase i s s hown b elow t he c ircle m ap.
