丹参不定根组织培养的研究(Ⅱ)

碳源、氮源和磷源对丹参不定根培养的影响

摘 要:目的 研究碳源、氮源和磷源对丹参S alvia miltior r hiz a不定根生长及其有效成分丹参酮ⅡA和原儿茶醛量的影响。方法 利用组织培养技术结合HP LC分析手段,通过改变培养基碳源种类和浓度、氮源和磷源浓度,研究其对丹参不定根生长和丹参酮ⅡA、原儿茶醛量的影响。结果 碳源、氮源、磷源对于不定根培养是必需的。以蔗糖为碳源,添加30g/L蔗糖,培养20d,不定根增殖倍数最高,60g/L蔗糖最适合丹参酮ⅡA合成,低质量浓度蔗糖更利于原儿茶醛合成。间歇添加蔗糖至培养25d,不定根增殖率是对照的2.3倍,丹参酮ⅡA量是对照的2.4倍。培养基N H4+和N O3-总浓度保持60mmo l/L,N H4+/N O3-为1∶4、1∶4、1∶1时分别得到最大的不定根增殖倍数、丹参酮ⅡA和原儿茶醛的量。改变培养基K H2P O4浓度时,丹参不定根生长均比对照组旺盛,但高浓度K H2PO4抑制了丹参酮ⅡA合成。结论 本实验确定了丹参不定根悬浮培养的最佳碳源种类和浓度、氮源比例和磷源浓度,表明不同碳源、氮源和磷源对丹参不定根生长和次生代谢物合成有显著影响。

Methods T he adv entitious r oots w ere obtained through tissue cultur e by m anipulation of carbon, nitro gen,and phosphate sources and the contents o f tanshinoneⅡA and pro to catechuic aldehy de were deter mined by HPLC.Results Car bon,nitr ogen,and phosphate so urces w er e necessar y fo r the cultur e of tior rhiz a adventitious r oots.T he highest times of r oot m ultiplicatio n w er e achiev ed at sucrose level of30g/L after20d culture,60g/L sucro se and low level sucrose w er e fav orable for biosyntheses of tanshinoneⅡA and pr otocatechuic aldehyde,respectively.The highest r oot y ield and tanshinoneⅡA co ntent o n day25w ere obtained by inter mittent sugar adding during cultivation,and the pr oduction of adventitious roo ts and tanshino neⅡA w ere2.3-and2.4-fold co mpared w ith those o f contro l,respectiv ely. The m aximum root grow th rate,contents of tanshinoneⅡA and protocatechuic aldehy de w ere achieved w hile NH4+-NO3-w as1∶4,1∶4,and1∶1,r espectively w hen concentration of total nitrog en sour ce w as kept at60mm ol/L.T o compare w ith the co ntrol g roup,chang ing of KH2PO4concentr ation co uld fav or for the adventilious r oot grow th,but high KH2PO4concentr ation inhibited tanshinoneⅡA biosynthesis. Conclusion T he results show that various carbon,nitrog en,and pho sphate sources have the significant effects on adv entitious r oot culture o f tior rhiz a.T he best carbo n source and its concentration, nitro gen and phosphate sources fo r the grow th of tior rhiz a adventitio us roo t and the synthesis of
