可靠的灭菌
AKTApilot 的灭菌效率经受了灭菌程序的测试，该法用 1 M 氢氧化钠 (NaOH) 作为杀菌剂进行微生物攻击实验。系统用包括由美国药典 United States Pharmacopoeia (USP 25) 推荐的三个细菌系和一种通常用于生产环境的酵母菌系的溶液感染系统 (表 1)。USP 25 要求菌落形成单位呈 log 6 减少。

流程 1. 样品阀 (V3) 选择一个合适的样品进口管道。样品泵 (P-908) 经过一快速响应的压力传感器，流动方向阀 (V5) 和柱选择阀 (V6+V7) 将样品溶液输送到柱中。可在进口管处用遥控的空气传感器 (2) 来检测样品容器是否腾空。 2. 进口阀 (V1+V2) 选择洗脱液进口管道。系统泵 (P-907) 经过一快速响应的压力传感器 (1) 和混合器将洗脱液输送到空气阱阀 (V4）。液体会被引导流过或绕过空气阱。 3. 经过空气阱阀 (V4) 后，洗脱液经过空气传感器 (1) 和压力传感器 (2) 流向流动方向阀 (V5）。空气传感器用于防止空气进入层析柱内。压力传感器监测柱头的压力。 4. 流动的液体继续经过其中之一的层析柱柱阀 (V6+V7) 进入柱内。液体流经层析柱的凝胶，样品被纯化。 5. 然后液体经过压力传感器 (4) 通过装在 pH 流动池的 pH 电极，电导池和 UV 流动池。柱前和柱后的压力传感器持续检测层析柱进口和出口的压力，屏幕上显示出不同的压力读数和变化中的压力曲线。 6. 液体继续流到出口阀 (V8+V9），出口阀用于转换液体到废液或分部收集器。

pH，电导和 UV 传感器 AKTApilot 配备有无菌设计的传感器以便连续地在线测量pH，电导和UV 吸收。检测器通过自我测试和自我校准提供准确和可靠的检测。这些流动池紧密地放在一起，这样减少了谱带的增宽和检测器之间的时间滞后。这些流动池很容易从面板上找到，方便维护。

如何选择AKTA？

AKTA explore 就正如Explore 这个单词的意思一样，探索，所以他是专门用来做工艺研发的; 1. 首先他的流速比Basic 要高，Basic 的流速0-10ml/min 而Expoler 100流速可以到0-100ml/min; 2. Explore 有7个柱位，可以如果想更换色谱柱，就必须把柱子先从机器上拆下来，然后再换,而Explore不需要； 3. Explore有Scouting的功能，能够设计一连串的实验，让机器自动运行，而Basic不具备Scouting的能力； 4. 他们的收集器也不相同，Explore 是Frac-950的收集器，而Basic最多就是Frac-920; Frac-950收集器可以收集10ml的管子，也可收集30ml的管子，而Basic 只能收集10ml的管子 5. Explore有自动配置缓冲液的功能，而Basic不能够自动配置缓冲液，所有的操作都得人为操作； 6. 最后一点就是他们两个价格不一样了，Basic可能30多万吧，而Explore可能要100万，价格差很多呦

A K蛋白纯化系统操作 Modified by JACK on the afternoon of December 26, 2020AKTA蛋白纯化系统操作AKTA蛋白纯化系统是当前蛋白纯化工作经常用到的一组设备，自动化程度很高。

AKTA系统依据不同的配置，可以分为AKTA EXPLORER、AKTA PILOT、AKTA PURIFIER等多种型号的设备。

以下以AKTA EXPLORER为例简单介绍AKTA蛋白纯化系统的一般操作。

1、认识AKTA。

AKTA explorer 是为方法开拓及研究应用而设计的全自动液相色谱系统。

该色谱系统的分离装置有三个主要组件，在底部平台的左侧整齐堆起（Fig 1）。

它们是：FIG 1、AKTA EXPLORER主机Pump-900 为双通道高效梯度泵系列。

在AKTAexplorer 100，流速范围ml/min，压力高达10 Mpa（泵名为P-901）。

在AKTA explore10，流速范围ml/min，压力高达25 Mpa（泵名为P-903）。

Monitor UV-900，同时监控190－700 nm 范围内高达3 个波长的多波长紫外-可见（UV-Vis）监测器。

（针对部分AKTA PURIFIER机型，尚有UPC-900监测器可供选择，光源为汞灯光源，一次可以监控一个波长，安装滤光片后，可以在选择的波长范围内进行切换。

）Monitor pH/C-900，在线电导和pH 监测的组合监测器。

柱被挂在装阀的门的外侧。

分离装置由UNICORN 软件控制。

软件安装于一独立的电脑主机之中，在电脑与色谱系统之间的通信由数据采集装置CU950进行控制。

2、一般操作开机按位于底部平台前左侧的ON/OFF 按钮，打开色谱系统，然后打开电脑电源。

二、水质分析仪使用注意事项：1.系统全密闭问题。

卡尔—费休试剂液路部分连接确定要紧固，从试剂瓶到计量泵再到反应池，否则发生试剂泄漏将直接影响测试结果。

其不密闭的另一个问题是测试时由于卡尔费休试剂在试验中吸取空气水分，会导致滴定尽头延迟。
