流程 1. 样品阀 (V3) 选择一个合适的样品进口管道。 样品泵 (P-908) 经过一快速响应的压力传感器, 流动方向阀 (V5) 和柱选择 阀 (V6+V7) 将样品溶液输送到柱中。 可在进口管处用遥控 的空气传感器 (2) 来检测样品容器是否腾空。 2. 进口阀 (V1+V2) 选择洗脱液进口管道。系统泵 (P-907) 经 过一快速响应的压力传感器 (1) 和混合器将洗脱液输送到 空气阱阀 (V4) 。液体会被引导流过或绕过空气阱。 3. 经过空气阱阀 (V4) 后, 洗脱液经过空气传感器 (1) 和压力 传感器 (2) 流向流动方向阀 (V5) 。空气传感器用于防止空 气进入层析柱内。压力传感器监测柱头的压力。 4. 流动的液体继续经过其中之一的层析柱柱阀 (V6+V7) 进入 柱内。液体流经层析柱的凝胶,样品被纯化。 5. 然后液体经过压力传感器 (4) 通过装在 pH 流动池的 pH 电 极,电导池和 UV 流动池。柱前和柱后的压力传感器持续 检测层析柱进口和出口的压力, 屏幕上显示出不同的压力 读数和变化中的压力曲线。 6. 液体继续流到出口阀 (V8+V9) ,出口阀用于转换液体到废 液或分部收集器。

A K蛋白纯化系统操作 Modified by JACK on the afternoon of December 26, 2020AKTA蛋白纯化系统操作AKTA蛋白纯化系统是当前蛋白纯化工作经常用到的一组设备,自动化程度很高。
AKTA系统依据不同的配置,可以分为AKTA EXPLORER、AKTA PILOT、AKTA PURIFIER等多种型号的设备。
以下以AKTA EXPLORER为例简单介绍AKTA蛋白纯化系统的一般操作。
1、认识AKTA。
AKTA explorer 是为方法开拓及研究应用而设计的全自动液相色谱系统。
该色谱系统的分离装置有三个主要组件,在底部平台的左侧整齐堆起(Fig 1)。
它们是:FIG 1、AKTA EXPLORER主机Pump-900 为双通道高效梯度泵系列。
在AKTAexplorer 100,流速范围ml/min,压力高达10 Mpa(泵名为P-901)。
在AKTA explore10,流速范围ml/min,压力高达25 Mpa(泵名为P-903)。
Monitor UV-900,同时监控190-700 nm 范围内高达3 个波长的多波长紫外-可见(UV-Vis)监测器。
(针对部分AKTA PURIFIER机型,尚有UPC-900监测器可供选择,光源为汞灯光源,一次可以监控一个波长,安装滤光片后,可以在选择的波长范围内进行切换。
)Monitor pH/C-900,在线电导和pH 监测的组合监测器。

柱被挂在装阀的门的外侧。
分离装置由UNICORN 软件控制。
软件安装于一独立的电脑主机之中,在电脑与色谱系统之间的通信由数据采集装置CU950进行控制。
2、一般操作开机按位于底部平台前左侧的ON/OFF 按钮,打开色谱系统,然后打开电脑电源。

二、水质分析仪使用注意事项:1.系统全密闭问题。
卡尔—费休试剂液路部分连接确定要紧固,从试剂瓶到计量泵再到反应池,否则发生试剂泄漏将直接影响测试结果。
其不密闭的另一个问题是测试时由于卡尔费休试剂在试验中吸取空气水分,会导致滴定尽头延迟。

1.PURPOSE目的
AKTA pilot层析系统为乙脑疫苗原液车间新装设备,安装于纯化间,主要用于乙脑病毒的分离纯化(凝胶过滤层析和离子交换层析)。
通过对AKTA pilot层析系统的验证,确认购买设备的技术指标、型号、性能及设计符合生产和GMP要求。
对设备安装过程进行确认,安装后进行试运行,以证明设备达到设计要求和说明书规定的技术指标,在此基础上进行设备性能验证,以离子交换层析试车,证明不仅能满足生产需要,而且符合工艺标准的要求并且所配制的溶液能够达到预期的质量要求。
2.EQUIPMENT DESCRIPTION 设备描述
AKTA pilot是为工艺开发,进行放大或缩小工艺实验,以及小规模生产而设计的全自动高效液相层析系统。
该系统具有纯化从几毫克到几十克产品的容量,并且是生物兼容,卫生的和可灭菌的系统。
AKTApilot符合药物开发中I-III期临床试验,试生产和正式生产的所有GLP和cGMP的要求。
(1)卫生处理的设计:有利进行无微生物和无污染的的安全生产。
(2)独有恒压调速设计:针对加样和装柱时的需要,可以在指定压力下,自动调控流速。
(3)高动态载量:流量为4-400 ml/min,梯度为0-100%;在限定的梯度下流速可达4-800ml/min。
(4)具有空气和压力传感器及四个进口的样品泵。
(5)无菌设计的内置分部收集阀,所有与液体有关的部件都是外置的。

7.2.SYSTEM STARTUP TESTING 系统启动确认
Objective目的:确认层析系统正常启动。
Acceptance Criteria可接受标准:UNICRN与层析系统通信连接成功,信号显示与说明和公司相关文件相符。
Test Procedure确认方法:
1.启动AKTA pilot电源开关,将电源开关由"O"扭至"I"位置;
2."POWER"绿灯闪烁,进入约1min的自检程序;
3.启动PC桌面""图标,输入用户名:“default”。
密码:“default”,确认进入;
4.打开“系统控制(System Control)”,选择“system:connect”将UNICRN与设备系统连接,点击"OK"。
5.当UNICRN与层析系统通信连接成功,显示一下信息:
