MACS CD4+ cells purification Kits:
1.Centrifuge cells at 300G×10min, Pipette off supernatant completely
2.Resuspend cell pellet in 40ul of buffer per 107 cells
3.Add 10ul of Biotin-Antibody Cocktail per 107 cells
4.Mix well and incubate for 10min at 40C
5.Add 30ul buffer per 107 cells
6.Add 20ul Anti-Biotin MicroBeads per 107 cells
7.Mix well and incubate for 15min at 40C
8.Wash the cells with buffer by adding 10-20×labeling volume and centrifuge at 300G×10min
9.Resuspend up to 108 cells in 500ul buffer.
10. Magnetic separation with LS columns
11. Place column in the magnetic field of LS MACS Saparator
12.Rinsing the column with buffer (LS: 3ML)
13.Apply cell suspension onto the column, collect effluent in a tube
14.Wash the column with 3ml buffer, 3 times,collect effluent in the same tube.
15.Count cells
