富硒金针菇菌种的选育

摘要

目的:食用菌以其独特的营养价值、药理作用和简易高效的生产工艺等优势备受青睐。金针菇(Flammnlina velutipes)作为食用菌中的一朵奇葩,更有美味可口、营养丰富、抗癌治溃疡等作用。目前对金针菇的营养成分报道较多,本文对金针菇菌丝体及其紫外诱变体的最佳富硒能力的培养基硒含量、菌丝生长特性、平板富硒培养与液体富硒培养、菌丝体硒含量等做了探讨。方法:本实验配置马铃薯综合培养基、紫外菌丝体诱变、富硒CM培养基培养金针菇201和白色金针菇;针对固体平板培养,采用十字交叉法测量菌落半径,使用乳酸石炭酸棉蓝染色液将菌丝染色,测其菌丝直径;针对液体发酵,使用游标卡尺测量菌丝球直径,收集菌丝球于称量瓶,在电热恒温干燥箱中烘干至恒重,用电子分析天平称其干重;使用紫外分光光度法测定菌丝含硒量;对实验数据的处理采用EXCEL制图、SPSS统计分析处理。结果:白色金针菇、金针菇201及其诱变菌种均能在含硒量为20~35μg/L的培养基上生长速度达到最快,富硒能力最强的状态。结论:相对于固体平板培养,液体发酵培养菌丝体的生长速度及富硒能力达到新的高度。

The Breeding of Selenium-accumulating Strain of Flammulina velutipess

Abstract

Subject: Edible fungi is extremely popular for its unique nutritional value, high-efficiency production process, and pharmacological effects. Flammulina velutipes, as a delicious and eutrophy fungi, also have the curative effect to anti-cancer and ulcers. At present, the nutrition constituent of Flammnlina velutipes is reported mostly, this research is about to discover the fittest one for mycelium of Flammulina velutipes and its UV-irradiation in selenium-accumulating media content, growth characteristics, differences between flat cultivation and cultivation of liquid selenium of selenium- accumulating mycelia, and selenium content of mycelium. Methods: This research configure potato integrated medium, ultraviolet radiation mutagenesis on mycelium, and Se- accumulating CM medium for training Flammnlina velutipes 201 and white Flammnlina velutipes. In the plate culture, I measure colony radius by cross cross method, lactic acid carbolic acid cotton blue dyeing liquid will be used in mycelium dyeing, measuring its mycelium diameter. In the liquid fermentation, calculate mycelium ball diameter by Vernier caliper. Mycelium ball in the bottle, drying to constant heavy in the electric thermostat dry box, calculate its dry heavy with Precision electronic balance. Using ultraviolet Spectrophotometric method determinates of selenium content of mycelium. The excel mapping and SPSS statistical analysis will be used in the processing of experimental data. Results: When selenium concentration of culture is 20~35 μ g/l, the fastest growth rate and selenium status of the most powerful of Flammnlina velutipes 201 and white Flammnlina velutipes and mutation strain will be achieved in the culture.Conclusions: Comparing with the plate culture, mycelium growth rate and capacity of Se-accumulating reaches new standard in the liquid fermentation.
