α-酮戊二酸受体1及Vimentin在糖尿病肾病大鼠肾脏的表达及意义

[摘要] 目的研究α-酮戊二酸受体1(oxoglutarate receptor 1, OXGR1)及Vimentin在糖尿病肾病大鼠肾脏的表达变化特点,并探讨其意义。方法选用Wistar大鼠,设置正常对照组和糖尿病模型组。通过注射链脲佐菌素,制作大鼠糖尿病模型;检测动物体重、血糖、24h尿蛋白及尿蛋白/尿肌酐比值的变化,并观察肾脏组织病理学的变化特点。应用免疫荧光检测OXGR1及Vimentin在肾脏的表达。结果正常对照组动物肾组织没有检测到OXGR1与Vimentin阳性重叠的区域。模型组动物随着病程的进展,体重减轻并出现血糖升高;24h尿蛋白及尿蛋白/尿肌酐比值升高,提示肾功能异常。免疫荧光发现模型组大鼠肾组织OXGR1或Vimentin阳性区域逐渐增强,同时两者阳性重叠区域逐渐增多。结论糖尿病模型动物随着病程的进展,一方面肾脏功能受损并出现组织学改变,另一方面OXGR1与Vimentin阳性重叠的区域增多,说明OXGR1可能与糖尿病肾脏上皮-间充质转化关系密切,前者在后者的诊断中具有潜在的意义。

Abstract: Objective To study the expression of oxoglutarate receptor 1 (OXGR1) and Vimentin in diabetic nephropathy, and its significance. Methods Wistar rats were randomly divided into normal control group and diabetes groups.Diabetes was induced in rats by using streptozotocin; body weight, blood sugar, urine protein in 24h and ratio of urine protein to creatinine in 24h were measured, and the pathological changes of kidneys were observed. Expression of OXGR1 and Vimentin in kidneys was detected by immunofluorescence(IF). Results In the normal control group, there was no OXGR1 and Vimentin overlapping region in the kidneys. In diabetes group, with the aggravation of diabetes, body weights of rats decreased, but blood sugar levels increased, and urine protein and ratio of urine protein to creatinine in 24h also increased, suggesting renal dysfunction; by IF, the OXGR1 or Vimentin positive regions gradually increased, and OXGR1 and Vimentin overlapping regions gradually increased, too. Conclusion With the progress of diabetes, the diabetic rats suffered from renal dysfunction, which was proved by pathological changes. Meanwhile, OXGR1 and Vimentin overlapping areas in the kidneys of diabetic rats increased, indicating OXGR1 may have a close relation with epithelial - mesenchymal transition in diabetic nephropathy, and the former might have potential significance in the diagnosis of the latter.
