K ey w ord s : soybean hu ll perox id ase ; crystallization ; purificat io n 交 联 酶 晶 体 ( Crosslinked Enzym e Crystals, CLEC s ) 是生物酶催化剂发展的一个里程碑, 与其 他形式的酶相比 , 其对温度、 p H、 和有机溶剂的稳定 [ 1 2] 性较高 。例如青霉素 G 酰 化酶的交联酶晶体 用于半合成头孢菌素前体的生产, 进行了 1000 批次 的反应后, 交联酶晶体仍 保持了 70 % 的初始活性。 酶作为生物大分子, 其结晶难度远高于一般的无机 小分子, 使酶结晶的成本很高 , 处理量很小。所以 , 研究新的结晶方法, 大批量低成本地生产酶晶体, 才 能使交联酶晶体实现工业应用。 常用的蛋白质结晶方法有 : 微池法结晶, 蒸汽扩 散法 , 液 液扩散法和透析结晶法等 。现有的 结晶方法结晶量小, 需要使用高度纯化的蛋白质, 常 [ 6] 使用离子交换或亲和 层析等方法进行 纯化 。常
