真菌论文:药用真菌桑黄化学成分的研究
【中文摘要】针层孔属真菌鲍氏针层孔菌(Phellinus baumii Pilat)属于锈革孔菌科(Hymenochaetaceae)、针层孔菌属(Phellinus Quel.),是我国传统药物桑黄的重要来源菌之一,素有“森林黄金”的美称。主要寄生于紫丁香属、李属、山楂属、等阔叶树干上。传统中药中,有抗肿瘤、保护肝脏、治疗胃溃疡等多种功效。为进一步开发我国珍贵的桑黄资源,本文对天然野生桑黄和人工培养桑黄95%乙醇提取物进行了系统的化学成分研究,共分离得到57个化合物。从天然桑黄中分离鉴定了50个化合物,其中有11个新化合物,包括4个新苄基二氢黄酮、2个新甾体、5个新苯乙烯基吡喃酮化合物,所有新化合物的结构是利用四大波谱技术联合解析得以确证的,包括红外技术(IR)、紫外技术(UV)、低分辨及高分辨质谱技术(MS)、核磁共振技术(NMR),并且利用到了异核单量子相关(HSQC)、异核多键相关(HMBC)、同核化学位移相关(COSY)等二维核磁共振技术(2D NMR)辅助新化合
物的结构解析。此外,利用高效液相-圆二色谱串联技术(HPLC-CD)证明两个苄基二氢黄酮分别是一对手性对应异构体的混合体,并且证明R、S构型在自然界是不等比例存在的。我们对这9个新多酚类化合物的NF-κB核转录因子抑制活性进行了评估,发现桑黄中多酚类化
合物对NF-κB核转录因子的确具有抑制活性,并且具有浓度依赖性。从人工培养桑黄中分离得到9个化合物,均是从该培养菌中首次分离得到。
【英文摘要】Phellinus baumii Pilat, a fungus belonging to the family Hymenochaetaceae, known as “Forest Gold”, which is one of the most important Chinese traditional medicinal Phellinus fungi, preferably hosts on the stems of genus Prunus, Syringa, Crataegus, and other hardwood trees. Its fruiting body has been traditionally used for treatment of gastrointestinal cancer, liver or heart diseases, and stomach ailmentsa in China.In orther to investigate our Chinese precious Phellinus fungi resource, we systematically study the chemical composition of phellinus fungus. In this paper,57 compounds were isolated from the EtOAc portion of the 95% ethanolic extract of the fruit body of P. baumii and the artificial culture of phellinus fungus. Especially,50 compounds were isolated from natural phellinus baumii fungus,11 of which were new compounds, including two pairs of epimeric benzyl dihydroflavone and five hispidin-derived polyphenols, phellibaumins A-E, along with two new sterols. The structures of new compounds were elucidated by spectroscopic methods including IR, MS, HRMS, 1D and 2D NMR experiments. In addition, with help of the HPLC-CD coupling technique, the two new dihydroflavones were verified to be a mixture of two enantiomers respectively, and we found these epimeric pairs
were non-equivalently present in nature. Inhibitory activities of phenolic compounds against NF-κ B in a dose-dependent manner have also been tested. From the culture of phellinus,9 compounds were identified, all of which were isolated from this aritifical fungus for the first time.
