产碱性蛋白酶菌株的筛选_分子鉴定及其酶学性质的初步研究

碱性蛋白酶是在碱性范围内水解蛋白质肽键的酶类,一般最适pH值为9~11,属内肽酶中的丝氨酸蛋白水解酶类,主要用于加酶洗涤剂中,在制革、纺织、医药、化妆品等的生产中也有广泛的应用[1]。目前,全世界所生产的酶中碱性蛋白酶占到总产量的30%左右,显示出良好的使用性能和经济价值。

碱性蛋白酶广泛存在于细菌、放线菌和真菌中,其中以芽孢杆菌属的碱性蛋白酶研究的最深入。本实验从山东东营的盐碱地采集土样,从中分离出若干株产碱性蛋白酶的菌株,并对其进行16S rRNA分子鉴定以及酶学性质的初步研究,以期为工业应用提供基础理论依据。

1材料与方法

1.1土样来源

山东东营盐碱地中采集土样。

1.2培养基

富集培养基:牛肉膏0.5g、蛋白胨1g、NaCl0.5g、水

摘要:从盐碱地土壤中筛选到5株产碱性蛋白酶的细菌,分别命名为TCCC11001、TCCC11004、TCCC11013、TCCC11024、TC-CC11029。将其革兰氏染色、16S rRNA鉴定为Bacillus subtilis、Bacillus alcalophilus、Bacillus pumilus、Bacillus subtilis、Bacillus licheni-formis。将菌株接入发酵培养基中,37℃、180r/min摇床培养48h,发酵液进行不同处理后Folin法测定酶活。结果发现:TCCC11001、TCCC11029的最适作用温度为40℃,TCCC11004、TCCC11013、TCCC11024的最适作用温度为50℃;TCCC11001、TCCC11004、TC-CC11013、TCCC11024、TCCC11029的最适作用pH值分别为10.0、11.0、10.0、9.0、11.0;TCCC11004、TCCC11013于40℃保温2h,残留酶活为最高酶活70%;TCCC11001、TCCC11024、TCCC11029的残留酶活<50%。DFP、PMSF完全抑制酶的活性,表明5种蛋白酶是典型的丝氨酸蛋白酶。

Screening,molecular classification of alkaline protease-producing strains and enzyme characteristics

Abstract:5strains which could produce alkaline protease were isolated from soil and named TCCC11001,TCCC11004,TCCC11013,TCCC11024 and TCCC11029.They were regarded as Bacillus subtilis,Bacillus alcalophilus,Bacillus pumilus,Bacillus subtilis,Bacillus licheniformis after deter-mination of16s rRNA and Gram staining.Enzyme activities were determined after incubating these strains at37℃,180r/min for48h.The results showed the optimal temperature of TCCC11001and TCCC11029was40℃,and for the others,the temperature was50℃;the optimal pH of these five strains were10.0,11.0,10.0,9.0and11.0respectively.Residual enzyme activities of TCCC11004and TCCC11013were as much as70%com-pared to the maximum activity when incubated at40℃for2h,and residual enzyme activities of TCCC11001,TCCC11024,TCCC11029were less than50%.The enzyme activity could be completely inhibited by DFP and PMSF,and it showed that these five proteases were typical serine protease.
