ABSTRACT: Objective To explore the dynamic changes of inflammatory factors expression at different time points during the treatment of acne with 5 -aminolevulinic acid photodynamic therapy (ALA -PDT) based on rabbit ear acne model. Methods A rabbit ear acne model was established by intradermal injection of Cutibacterium acnes (C.acnes) into the skin of rabbit ears. The successfully established acne lesions were treated with ALA-PDT. HE staining was used to evaluate the variation trend of tissue inflammatory response. RNA was extracted from rabbit ear tissue, and real-time PCR was used to detect the relative expression levels of inflammatory factors such as IL-1琢, IL-1茁, IL-6, IL-8 and NF-资B (p65) mRNA before and at 4, 24 h and 7 days after ALA-PDT treatment. Results By evaluating the HE staining, it was found that the inflammation performance of skin lesion aggravated gradually at 4-24 hours after ALA-PDT treatment, and the infiltration of inflammatory cells decreased significantly at 7 days after ALA-PDT treatment. Real-time PCR showed that the relative expression levels of IL-1琢,IL-1茁 and IL-6 mRNA increased at 4 and 24 hours after ALA-PDT treatment compared with those before treatment, but the differences were not statistically significant (P>0.05), while the relative expression levels of IL -8 and p65 mRNA increased significantly, and the differences were statistically significant (P <0.05); the relative expression levels of all inflammatory factors decreased significantly at 7 days after ALA -PDT treatment compared with those before treatment, and the differences were statistically significant (P<0.05). Conclusion ALA -PDT may amplify inflammation by up -regulating IL -8 and p65 in the early stage, then inhibits inflammatory factors to improve tissue inflammation, and finally effectively treat acne.
