利用液相色谱法测定中药的柚皮苷含量

摘要】目的建立HPLC法测定常见中药中柚皮苷的方法，测定常见中药中柚皮苷含量。方法 Waters Symmetry C18柱（4.6x150mm,3.5μm），流动相为甲醇:乙腈:磷酸缓冲液（0.01mol/L pH3.5）=9:3:8，检测波长为280nm。结果柚皮苷的出峰时间为2min，柚皮苷1.80～350mg/L范围内具有良好的线性关系，R2值为

0.9999，平均回收率分别为99.5%-102.5%，相对标准偏差小于5%；香橼、枳实和橘红的柚皮苷含量分别为2.89-5.67%、0.21-0.26%、11.97-2.44%，不同产地的香橼、枳实、橘红中柚皮苷含量的相对标准偏差分别为28.88%、10.63%和26.42%;四磨汤、胃痛丸、橘红丸颗粒和抗骨增生丸中柚皮苷的含量分别为5.99mg/g、2.25mg/g、1.71mg/g、0.30mg/g，不同厂家生产的四磨汤、胃痛丸、橘红丸颗粒和抗骨增生丸中柚皮苷浓度的相对标准偏差分别为8.43%、14.72%、9.90%和

3.42%。结论建立了一种简便、准确、适用性广泛地测定中药中柚皮苷的方法，产地对香橼及橘红中柚皮苷含量的影响较大，而不同厂家生产的中成药中柚皮苷的含量偏差相对较小。

Determination of naringin in Chinese medicine byhigh-performance Liquid Chromatographic Method

【Abstract】Aim: To set up a quantitativehigh-performance liquid chromatographic method for determination of naringin and to investigation naringin content in common Chinese medicine. Method: An RP-HPCL method was set up, using Symmetry C18 column, methyl alcohol: acetonitrile:PBS(containing 0.01mol/L postassium dihydrogan phospate pH=3.5)= 18:3:14 as mobile phase with a flow rate of 1.0ml/min at the detection wavelength of 210nm. Result: retain time was about 2min, naringin well separated with other compositions. The Calibration curve was ofgood linearity in the range of 1.80～350 mg/L with R2 0.999, recoveries were range from 99.5%-102.5%, relative standard derivation was less than 5%. Naringin content in Citron, Citrus aurantium, and orange-red were 2.89-5.67%, 0.21-0.26%, and 11.97-2 .44%, respectively. Relative standard derivation of naringin contents from different origin were 28.88 for citron, 10.63 for citrus aurantium, 26.42 for orange-red. Naringin content in 4 soup, stomach pain pill, orange pill bone particles and anti-proliferative Pill were 5.99 mg /g, 2.25mg /g, 1.71mg /g, and 0.30mg /g repectively. Relative standard derivation of naringin contents from different factory were 8.43 for 4 soup, 14.72 for stomach pain pill, 9.90 for orange pill bone and 3.42 for anti-proliferative pill. Conclusion: This method is accurate, sensitive and convenient for detecting naringin content of Chinese medicine, Origingreatly effects naringin content of the citron and orange-red ,whereas, naringin content of Chinese patent medicine are little affected by origin.
